About this Role:

The Kansas State Veterinary Diagnostic Laboratory (KSVDL) within the College of Veterinary Medicine is seeking applicants for a Research Assistant in the Virology Laboratory. The duties of this position include performing tests on various analytes such as serum, fetal fluid, and tissues using Serum Neutralization (SN), Hemagglutination Inhibition (HI), Virus Isolation, Enzyme Linked Immunosorbent Assay (ELISA) and Immunoflourscence Assay (IFA) or other tests to detect titers, abnormalities, or disease and enter findings into current Laboratory Information Management System. Set up, adjust, maintain and clean laboratory equipment and maintain the quality system through document reviews, trend analysis for all tests as well as corrective action reports and ensuring the quality of test results.

Duties / Responsibilities:

 

• Learn and perform techniques listed below or as requested by section head/laboratory supervisor.

SN (Serum Neutralizations):

 

• Make serial dilutions of test samples and controls on a 96-well plate

• Prepare stock virus, including obtaining virus, proper dilutions of stock for test, setting up back titrations

• Make cell culture suspension using appropriate cell line and concentration for each test

• Heat inactivation of samples and creation of worksheets for SNs scheduled to be run

 

HI (Hemagglutination Inhibition):

 

• Make serial dilutions of test samples and controls on a 96-well V bottom plate

• Prepare stock virus, including obtaining virus, proper dilutions of the stock for test, setting up back titrations

• Make blood suspension using appropriate blood type and concentration for each HI test

• Heat inactivation of samples and creation of worksheets for HI scheduled to be run

• Prepare sample set up using correct concentration of PBS (Phosphate Buffered Saline) and blood ELISA:

• Perform Antigen and Antibody ELISAs according to approved protocols or manufacturers' specifications

 

IFA:

 

• Make and/or have in stock plates and slides

• Dilute samples appropriately

• Use appropriate FITC conjugate

• Read using fluorescent microscope

 

CELL CULTURE:

 

• Perform daily maintenance of cell cultures including feeding and splitting

• Make cultures for procedures in other laboratories

• Freeze down cell stock and maintain liquid nitrogen log

 

GENERATION OF ASSAY STOCKS:

 

• Culture laboratory strain viruses for assay stocks by inoculating into cell cultures

• Harvest viral fluids and perform titrations to determine the titer of the stock

• Aliquot appropriate volumes of virus, label vials, and record stock information for quality assurance purposes

 

VIRUS ISOLATION:

 

• Process tissue samples and prepare other clinical specimens for inoculation

• Perform basic VI's with prepared specimens, inoculating samples into seeded culture flasks or plates

• Monitor cultures for viral growth through light microscopy

• Identify viral pathogens through fluorescent antibody procedures

 

LABORATORY INFORMATION MANAGEMENT:

 

• Assign sample identity

• Order tests

• Create results worksheets

• Review results

• Finalize test results

 

QUALITY SYSTEM:

 

• Perform quality assessment and quality control documentation needed for the Lab to stay in compliance with regulations

• Assist in SOP updates

• Corrective actions

• Perform other duties assigned by sections head or supervisor in the lab

 

ORDERING/CUSTOMER SERVICE:

 

• Keep supply inventory

• Order items as needed according to approved budget

• MAS200

• Assist customers via phone, email

 

Required Minimum Qualifications:

 

• Bachelor of Science in Virology, Microbiology or related field

 

Preferred Qualifications:

 

• At least two years of laboratory experience

• Excellent interpersonal, organizational and communication skills
